Marvel Studios gradually revealed the main cast of Avengers: Doomsday in March 2025 through names on director's chairs. During the five-hour presentation, Tom Holland, Benedict Cumberbatch, Brie Larson, Mark Ruffalo, and Jeremy Renner were not included in the list of 27 lead actors.
The post-credits scene of Spider-Man: Brand New Day hints that Spider-Man will travel to outer space, but does not confirm his participation in Avengers: Doomsday. The film also leaves questions about Hulk's subsequent whereabouts, and new footage in the re-release provides further answers: Bruce Banner was transferred to an underground military base claimed to be impenetrable, but someone subsequently breached it.

Owen Wilson, Ke Huy Quan, and Eugene Cordero reprise their roles as Mobius, O.B., and Casey, respectively, with Tara Strong continuing to voice Miss Minutes. This will mark the first appearance of these four characters in a theatrical Marvel Cinematic Universe film.
The Time Variance Authority first appeared in the series Loki, responsible for handling anomalies in the timeline, and later appeared in Deadpool & Wolverine. In the re-release clips, Mobius and others witness the destruction of multiple Earths tied to Marvel Cinematic Universe history, and the four characters are expected to address the crisis facing surviving heroes and refugees in Avengers: Doomsday.
Sylvie, who has close ties to these Time Variance Authority members, has not been confirmed to return. There is currently speculation that she may be connected to the four Latverian witches recruited by Doctor Doom, but actress Sophia Di Martino has only liked related questions without providing a response.
